Ørtenblad, Niels, Gisela Sjøgaard, and Klavs Madsen. Impaired sarcoplasmic reticulum Ca release rate after fatiguing stimulation in rat skeletal muscle. J Appl Physiol 89: 210–217, 2000.—The purpose of the study was to characterize the sarcoplasmic reticulum (SR) function and contractile properties before and during recovery from fatigue in the rat extensor digitorum longus muscle. Fatiguing contractions (60 Hz, 150 ms/s for 4 min) induced a reduction of the SR Ca release rate to 66% that persisted for 1 h, followed by a gradual recovery to 87% of prefatigue release rate at 3 h recovery. Tetanic force and rate of force development (1dF/dt) and relaxation (2dF/dt) were depressed by ;80% after stimulation. Recovery occurred in two phases: an initial phase, in which during the first 0.5–1 h the metabolic state recovered to resting levels, and a slow phase from 1–3 h characterized by a rather slow recovery of the mechanical properties. The recovery of SR Ca release rate was closely correlated to 1dF/dt during the slow phase of recovery (r 5 0.51; P , 0.05). Despite a slowing of the relaxation rate, we did not find any significant alterations in the SR Ca uptake function. These data demonstrate that the Ca release mechanism of SR is sensitive to repetitive in vitro muscle contraction. Moreover, the results indicate that 1dF/dt to some extent depends on the rate of Ca release during the slow phase of recovery.

Reduced expression of sarcalumenin and related Ca2+ -regulatory proteins in aged rat skeletal muscle.

In skeletal muscle, Ca(2+)-cycling through the sarcoplasm regulates the excitation-contraction-relaxation cycle. Since uncoupling between sarcolemmal excitation and fibre contraction may play a key role in the functional decline of aged muscle, this study has evaluated the expression levels of key Ca(2+)-handling proteins in senescent preparations using immunoblotting and confocal microscopy. S...

Studies on the in Vitro Interaction of Electrical Stimulation and Ca++ Movement in Sarcoplasmic Reticulum

Sarcoplasmic reticulum fragments (S.R.F.) were isolated from skeletal and heart muscles. These fragments were found to take up Ca(++) very actively from media. When monophasic square waves were passed through the S.R.F. suspension, the Ca(++) uptake by S.R.F. was decreased. When the suspension was stimulated electrically after the Ca(++) was taken up by S.R.F., the initiation and the cessation ...

Glycogen and glycogen phosphorylase associated with sarcoplasmic reticulum: effects of fatiguing activity.

The purpose of the present study was to investigate the effects of fatiguing muscular activity on glycogen, glycogen phosphorylase (GP), and Ca(2+) uptake associated with the sarcoplasmic reticulum (SR). Tetanic contractions (100 ms, 75 Hz) of the gastrocnemius and plantaris muscles, elicited once per second for 15 min, significantly reduced force to 26.5 +/- 4.0% and whole muscle glycogen to 2...

Modulation of sarcoplasmic reticulum Ca2+ release in skeletal muscle expressing ryanodine receptor impaired in regulation by calmodulin and S100A1.

In vitro, calmodulin (CaM) and S100A1 activate the skeletal muscle ryanodine receptor ion channel (RyR1) at submicromolar Ca(2+) concentrations, whereas at micromolar Ca(2+) concentrations, CaM inhibits RyR1. One amino acid substitution (RyR1-L3625D) has previously been demonstrated to impair CaM binding and regulation of RyR1. Here we show that the RyR1-L3625D substitution also abolishes S100A...
